Common in embryonic development and inflammatory responses. Often, the part of the system that is degraded by proteolysis is a transcription factor, so these pathways are common regulators of gene expression.
We will look closely at four examples:
Overview: Hedgehog is the signal ligand, which binds to a receptor complex that includes an inhibitor of signaling called “patched”. In the absence of hedgehog, patched is preventing an activator called “smoothened” from reaching the plasma membrane and a transcription factor called Cubitus interruptus (Ci) is degraded. The Ci degradation product acts as a negative regulator of gene transcription. Below are the steps that lead to the release of the cell from these negative inhibitory effects, and the production of a full length, active Ci protein.
